During this time Peter Weyland emerges. He is brilliant, cold, and ruthless. Take the genius of Steven Hawking, mix in Steve Job's charisma, and add the negative traits of todays CEOs (Bill Gates, Robert Murdoch, Larry Ellison).
The company he creates is called Weyland Industries. He not only grows the company with his inventions, but he lies, steals, and cheats into any sucess that escapes him. Nothing is permitted to interfere with achieving his goals.
Over the years he gains monopolies in one sector after another through manipulation and political trickery. Through constant mergers he soon controls defense, energy, health, and other countless other critical industries. While governments remain Peter Weyland is very much in control. The company builds the star ships. They make all the armies weapons. They provide eletricy, media, almost everything. Waring with Weyland Industries countires under control of asian rival Yutani Corporation provides distraction for the masses.
Problem is Peter Wayland is very much human and he's getting very old and sick. Now 83 he's used technology to live longer than his failing body should allow him to.
Desperate. He finds his answer in some archelogical discoverys. They indicate at some time in the distant past we were visited (if not created) by alien lifeforms and they left the locals with a map of what could be their home world.
From here you can tell they visited the alien world. My best guess is the aliens created man in their image, as they have similar features. They may have created all life on earth. As this likely occured thousands of years ago, the aliens must have an extremely long life span. Their technology rooted in biological engineering and bio mechanical constructs.
Some time during their exploration of the alien base (inside their ship sits), something goes horribly wrong. I'm guessing Peter Weyland (or those loyal to him) direct the crew into doing something unwise in his eagerness to have the alien technology for his own. Looks like some people get mutated and a bunch of bad things happen.
This likely pisses off the alien (or aliens). Being the creator of life, they have also created tools to destroy it. Including a sealed bays of thousands xenomorph eggs. Depsoited on earth, over the course of months they would make short work of the human species. I'm sure the alien give some sort of speech or indication on whats about to go down.
Realizing they are screwed the crew rams the alien space craft as it attempts to leave for earth. I wouldn't be surprised if this totally destorys their own ship.
It's unclear what happens from here. Perhaps the alien ship uses its ability to bend space/time to move to another planet (LV-426) where the last alien dies from a chestburster. I wouldn't be surprised if everyone dies.

Im seeing this as kind of the same thing that happens in "Mission to Mars". The humans are drawn to this planetoid that is being terraformed. There, they find the remnants of lost civilization, and a dormant Space Jockey ship. As it turns out, the SJ are actually advanced humanoids who have seeded the galaxy and then wait for their children to seek them out. It coukd be the SJ race wants to commune with their children by transforming them into "the mother race". Unfortunately, by "securing" a few of the ampules, they have set off a sort of fail-safe that sets in motion a chain of events that will lead to the destruction of humans on Earth.
One of the crew is "infected" by the DNA of an SJ, and begins to transform into one. His/her directive is to destroy the mission, secure a load of proto-alien lifeforms that are now emerging from the infected crew, and pilot the Space Jockey ship to earth. There, the protoforms will destroy all life and return the planet to it's pre-life state. In the scene where you see the SJ in the Pilot Seat, you see the bio-suit coming together around the transformed human. Now that the transformation is complete, his DNA interacts with the bio-mechanical controls and he can pilot the ship. He takes off.
The last survivors of Prometheus realize they have to stop this. They rig the ship to ram the SJ ship, which we see happen in the trailer. We also see the ship as it crashes. In the aftermath, the rest of the Prometheus crew dies, one way or the other, and the former human Space Jockey is infected by the protoforms it was to ferry to Earth.
The Alien tie-in - this planetoid IS LV-426. At the time Prometheus landed, it was in the midst of transformation from a dead world to a living planet. The SJ ship and the ampules were acheiving this purpose. After the demise of Prometheus and the Alien ship, the world reverted to it's primordial dead state - "primarily methane, deep cold - well below the line".
In a final act, as his/her humanity fades and the Space Jocky DNA takes over, a warning recording is programmed in the Space Jockey ship...

how long after ....did the nostromo set down on LV-426 prior to prometheus
and i think the idea of the whole crew being infected by airborne spores is the way to go !!
and i think ....humans and the other space craft land there at the same time ...not knowing what is happening ....and they are more advanced and try to save the human race !!
