I think that particular remark in the TED speech has hidden connotations. Could Weyland have gone through some unspeakable rite of passage to attain a certain level of existence or state?

Either that, or he is referencing the pain that comes with breaking a new barrier. Exploration always has risk and loss...

I think he's referring to a state of inner being where external pain isn't very important, hence his arrogant little snigger afterwards.

Probably more like "no pain - no gain". Nothing worthwhile was ever accomplished without some mild discomfort. Not referring to some super secret Engineer hazing ritual, though.
