Yes, it was after the head exploded. They were undoubtedly talking - in one way or another - about mission progress, and David was receiving orders.

One thing maybe is that David examined that urn full of goop so he might imply his examining it, like premature because he doesn't know what is inside.

Yeah thats what I think was going on. He was telling Weyland that maybe he doesnt know enough about the black goo or whatever they were specifically talking about. Maybe Weyland was trying to figure out if he knew anything and basically he was saying Eh its a bit premature to know exactly what this black goo is or can do. Those are just my thoughts. Cant wait for the bluray to come out and examine the film alot closer :D

I am pretty sure Weyland was asking him to test the goo on someone to see what it does. David picked Holloway simply because, in the words of M.Fassbender; "He was mean to me".
