Beyond discovering cave paintings/artifacts from around the world depicting giant beings pointing to a particular constellation, what other evidence did they come across to make them believe the Engineers created/seeded Humanity? Did they see the prologue like we the audience?? ( As an additional note, The hologram depicting the Lid of Palenque is wrongly labelled as being from Teotihuacan, Mexico, when it is clearly not. )

Didn't they do a DNA test on the alien head and match the DNA

Yes, the DNA provided the link between humans and engineers. But having said that, it doesn't really prove the engineers created humans.

The DNA analysis came afterwards. Earlier in the movie as they are explaining to the others the purpose of their mission, they adamantly state that we, humanity were created by them. They must have based that on something, even before they embarked on the mission on behalf of Weyland. And yes, a DNA match may only mean we have a common ancestor.
